The Effect of Plant Growth Regulators (PGRs) on Efficient Regeneration of 12 Recalcitrant Indica Rice (Oryza Sativa L.) Genotypes

Biotechnological approaches provide an efficient and expeditious system for developing agronomic features of rice at the cellular molecular level compared to traditional breeding methods. The in-vitro regeneration method optimization recalcitrant indica genotypes is a critical step biotechnological further development as optimum concentrations Plant Growth Regulators (PGRs) vary from genotype genotype. Here, 12 important genotypes, including local cultivars varieties, were collected different regions Bangladesh evaluated their potentiality. Mature dehusked sterilized seeds inoculated on basal media (MS, N6, B5) supplemented with 2,4-Dichlorophenoxyaceitic acid (2,4-D) callus induction. Induced calli transferred N6 combinations 6-Benzylaminopurine (BA) 1-Napthaleneacetic Acid (NAA) shoot Regenerated shoots rooted half-strength MS without supplementation any hormones. Treatments plant growth regulators showed varying results. Maximum induction frequencies observed 2.5-3.0 mg/l 2,4-D where Saat, Jamaibabu Nimikhanai highest (84.44%; 78.89 80% respectively) maximum initiation was 2.5 BA +1.0 NAA 3.0 6-BA +1.5 BRRI Dhan71, Nayapaijam, Dhan28 frequencies-86.67, 85.00, 86.67 88.33% respectively. Nearly all induced root formation media. This study demonstrates variations in response Jamaibabu, Nayapaijam potential.
